Description

This Colorimetric Cell-Based ELISA Kit from Assay Genie is designed to detect phosphorylated ABL1 at Tyr245 in cell lysates. It is a lysate-free, high-throughput and sensitive assay that uses an indirect ELISA format, in which ABL1 (Phospho-Tyr245) is captured by phospho-specific primary antibodies while HRP-conjugated secondary antibodies catalyze a colorimetric reaction measured at 450 nm. The kit is reactive with human, mouse and rat samples and is suitable for adherent cells, with a dynamic range of > 5000 cells. It can be used to measure the relative amounts of phosphorylated ABL1 in cultured cells and to screen for the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on ABL1 phosphorylation.

ABL1 is a key protein in cell signaling and regulation, contributing to cell proliferation, differentiation and survival, and phosphorylation at Tyr245 is important for its kinase activity. Dysregulation of ABL1 signaling has been implicated in cancer and other diseases, making the kit relevant for studying ABL1 phosphorylation in disease progression and potential therapeutic interventions.

Because the Cell-Based ELISA is qualitative, two normalization methods are provided. A monoclonal anti-GAPDH antibody serves as an internal positive control for normalizing target absorbance values, and the Crystal Violet whole-cell staining method can be used to determine cell density, allowing absorbance values to be normalised to cell number to adjust for plating differences.
