Description

This cell-based ELISA kit from Assay Genie is designed for the qualitative measurement of ABHD9 protein expression in cultured cells. It can be applied to adherent cells and works in a lysate-free format, enabling high-throughput screening of the relative effects that treatments, inhibitors (such as siRNA or chemicals) or activators have on ABHD9 levels in cell lysates and culture supernatants.

Detection is colorimetric, read at 450 nm, with a dynamic range of > 5000 cells, and the assay is configured for human reactivity. Qualitative determination is achieved through an indirect ELISA format in which ABHD9 is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyse a colour reaction on substrate addition. Because the readout is qualitative, two normalisation methods are supported: a monoclonal anti-human GAPDH antibody included as an internal positive control, and a Crystal Violet whole-cell staining step for adjusting results to cell density.

ABHD9 (epoxide hydrolase 3, gene symbol EPHX3, UniProt Q9H6B9) is a member of the AB hydrolase superfamily and the epoxide hydrolase family, with a molecular weight of 40,909 Da. It is described as a key enzyme in lipid metabolism with links to metabolic disorders and neurodegenerative diseases, placing the kit in the metabolism research area. The kit is stored at 4 °C with a stability of 6 months.
