Description

From Assay Genie, this colorimetric cell-based ELISA kit provides a sensitive, lysate-free, high-throughput method for detecting ABHD7 protein expression directly in adherent cells. The assay uses an indirect ELISA format in which ABHD7 is captured by target-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody and catalyzing a colorimetric reaction upon substrate addition, measured at 450 nm. The kit is reactive with human and mouse samples and is suited to measuring relative ABHD7 levels in cultured cells as well as screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on ABHD7 expression.

A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density and adjust for plating differences. The kit is supplied in a 96-well microplate format with a stated dynamic range of > 5000 cells, and is stored at 4 °C with a stability of 6 months.

ABHD7 (also designated EPHX4, Epoxide hydrolase 4, Abhydrolase domain-containing protein 7, and Epoxide hydrolase-related protein) is an integral membrane protein belonging to the AB hydrolase superfamily and the epoxide hydrolase family. It plays a key role in lipid metabolism and has been implicated in metabolic disorders, neurological disorders, and cancer, making accurate measurement of ABHD7 levels a useful tool for research into disease pathology and the development of targeted therapies.
