Description

A competitive ELISA, coated with antigen, designed by Assay Genie for the quantitative measurement of 6-Keto-PGF1α (6-Keto-Prostaglandin F1α), a stable metabolite of prostacyclin (PGI2). Because 6-Keto-PGF1α levels are widely used to evaluate prostacyclin turnover, the kit supports research into oxidative stress, inflammation, cardiovascular disease, thrombotic disorders, and related inflammatory conditions.

Validated for serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples, the colorimetric assay delivers a sensitivity of 9.375 pg/mL and a working range of 15.625-1000 pg/mL. Stored sealed at 2-8°C, the components must not be cryopreserved. Linearity across 1:2, 1:4, and 1:8 dilutions spans 85-97%, 90-104%, and 85-104% for serum (n=10); 84-101%, 91-101%, and 83-101% for EDTA plasma (n=10); and 81-94%, 80-98%, and 80-94% for heparin plasma (n=10). Recovery averages 94% (range 85-100%) for serum, 94% (range 85-104%) for EDTA plasma, and 97% (range 86-104%) for heparin plasma.

The protocol begins with a 30-minute TMB substrate equilibration at room temperature, followed by addition of 50 µL standard or sample and 50 µL biotin-labeled antibody per well, a 45-minute incubation at 37°C, and five washes with one-minute soak times. TMB substrate is then added and incubated in the dark at 37°C for 10-20 minutes, the reaction is stopped (color changes from blue to yellow), and absorbance is read at 450 nm using a microplate reader.
