4-Hydroxynonenal (4-HNE) is the principal α,β-unsaturated aldehyde generated when reactive oxygen species attack omega-6 polyunsaturated fatty acids in cellular membranes. This reactive lipid-peroxidation product forms covalent adducts with cysteine, histidine, and lysine residues of proteins, and its accumulation is implicated in neurodegeneration, atherosclerosis, alcoholic liver disease, insulin resistance, and tumor progression.
