14-3-3 zeta/delta (Phospho-Thr232)Colorimetric Cell-Based ELISA Kit

A cell-based ELISA kit from Assay Genie designed to measure 14-3-3 zeta/delta protein phosphorylated at threonine 232 in cell lysates. The assay uses a colorimetric detection method read at 450 nm and is intended for adherent cells, with reactivity confirmed for human, mouse and rat samples. It supports a dynamic range of > 5000 cells and provides a lysate-free, high-throughput workflow.

The kit follows an indirect ELISA format in which phospho-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step is provided to determine cell density, allowing absorbance values to be normalized to cell amounts. The assay targets the YWHAZ gene product (UniProt P63104, Gene ID 7534), a 14-3-3 family adapter protein implicated in cell signaling, cell cycle regulation, apoptosis and cell growth, with phosphorylation at Thr232 recognised as a regulator of its function.
