Description

This in-cell ELISA kit from Assay Genie is designed for the precise measurement of phosphorylated 14-3-3 beta/zeta at Ser184/186 in cell lysates and tissue homogenates. Using a colorimetric detection method read at 450 nm, the cell-based assay employs an indirect ELISA format in which target-specific primary antibodies capture the protein of interest and HRP-conjugated secondary antibodies catalyse the colorimetric reaction. The kit is reactive with human, mouse and rat samples and supports a dynamic range of > 5000 Cells, offering a convenient, lysate-free and high-throughput approach to characterise 14-3-3 beta/zeta phosphorylation and expression in cultured adherent cells.

Phosphorylation of specific serine residues (Ser184/186) on 14-3-3 beta/zeta is known to modulate its activity, making it a valuable biomarker for studying intracellular signalling pathways involved in cell cycle regulation, apoptosis and signal transduction. The assay can be used to measure the relative amounts of phosphorylated 14-3-3 beta/zeta in cultured cells and to screen for the effects of various treatments, inhibitors (e.g., siRNA or chemicals) or activators on its phosphorylation. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalising target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density and adjust for plating differences.
