Description

This colorimetric cell-based ELISA kit from Assay Genie is designed for the qualitative detection of 14-3-3 beta/zeta protein in cells. The assay uses an indirect ELISA format in which the target protein is captured by specific primary antibodies and detected with HRP-conjugated secondary antibodies, producing a colorimetric readout at 450 nm. It is described as a convenient, lysate-free, high-throughput and sensitive tool for measuring relative 14-3-3 beta/zeta levels in cultured cells, and for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on 14-3-3 beta/zeta expression.

The kit shows reactivity with Human, Mouse and Rat samples and is suitable for use with adherent cells, cell lysates and tissue homogenates. A dynamic range of > 5000 Cells is reported. Normalisation is supported by an included anti-GAPDH monoclonal antibody, which serves as an internal positive control, together with a Crystal Violet whole-cell staining step for determining cell density. The kit is supplied in a 96-well microplate format and is stored at 4°C with a stated stability of 6 months.

14-3-3 beta/zeta is a member of the 14-3-3 family of adapter proteins implicated in cell cycle regulation, signal transduction and apoptosis, and is encoded by the YWHAB/YWHAZ gene symbols. Dysregulation of 14-3-3 beta/zeta has been linked to cancer, neurodegenerative disorders and cardiovascular diseases, making it a useful biomarker in disease and cell-signalling research.
