Description
STEMCELL Technologies offers a maintenance kit designed for the feeder-free culture of human embryonic stem cells (hESCs) and human induced pluripotent stem cells (hiPSCs). The formulation is based on the E8 medium developed by the laboratory of Dr. James Thomson at the University of Wisconsin-Madison, and it contains only the essential components required to maintain pluripotent stem cells, providing a simpler alternative to more complex formulations.
The kit is referenced in protocols for thawing, passaging, and cryopreserving established induced pluripotent stem cell lines, including those adapted from feeder-dependent conditions and CRISPR/Cas9-edited lines. Cells are typically maintained as aggregates on a vitronectin matrix, using either full-length xeno-free or truncated protein, and may be cultured with or without Penicillin-Streptomycin in the media.
As a feeder-free, animal component-free system, this medium is suitable for downstream applications such as differentiation and CRISPR screens. It may be combined with either Corning Matrigel hESC-Qualified Matrix or Vitronectin XF as the culture matrix for a defined xeno-free workflow.
