STEMCELL Technologies presents a xeno-free medium designed for the in vitro culture and expansion of human hematopoietic cells when the appropriate growth factors and supplements are added. The medium contains pre-tested human-derived and recombinant human proteins, with material derived from human plasma. Donors have been tested and found negative for hepatitis B surface antigen (HBsAg) and HIV-1 antibodies and/or HIV-1 antigen; however, the product should be considered potentially infectious and handled using universal precautions.

When combined with the appropriate StemSpan expansion supplements, the medium can be used to expand CD34+ cells isolated from human cord blood, mobilized peripheral blood, or bone marrow samples, and to expand and differentiate lineage-committed progenitor cells into populations of erythroid, myeloid, or megakaryocyte progenitor cells. Compatible StemSpan supplements include CD34+ Expansion Supplement (10X), CC100, CC110, Erythroid Expansion Supplement (100X), Megakaryocyte Expansion Supplement (100X), Myeloid Expansion Supplement (100X), and Myeloid Expansion Supplement II (100X).

To prepare complete medium, thaw at room temperature (15 - 25°C) or overnight at 2 - 8°C and mix thoroughly. If not used immediately, aliquot and store at -20°C; do not re-freeze after thawing. Added cytokines, growth factors, and other components, along with cells in sterile cell culture medium, should not exceed ~10% of the total volume. Cultures are incubated at 37°C and 5% CO2, with a 7-day culture period described as optimal for cell yield, CD34 expression, and progenitor cell function. The product is stored at -20°C and remains stable until the expiry date on the label.
