StemSpan-AOF, 1 L Bottle

An animal origin-free and xeno-free hematopoietic cell expansion medium from STEMCELL Technologies is supplied as a 1000 mL ready-to-use liquid in a single bottle. StemSpan-AOF is built on a Phenol Red-free Iscove's MDM base and supplemented with recombinant human albumin, recombinant human insulin, recombinant human transferrin, and 2-mercaptoethanol, with no serum and no human- or animal-derived components. The medium is manufactured and tested under relevant cGMPs for cell therapy, gene editing, and immunotherapy research workflows.

In published validation data, human cord blood-derived CD34+ cells cultured in this medium for 7 days achieved approximately 20-fold expansion while maintaining the primitive CD34brightCD90+CD45RA- hematopoietic stem cell subset. Cells expanded under these conditions showed comparable or greater repopulation than uncultured controls when transplanted into sub-lethally irradiated NSG mice, measured at both short-term (3 week) and long-term (20 week) engraftment timepoints.

The defined, xeno-free formulation is intended for in vitro research use only and pairs with STEMCELL Technologies' matched supplements to drive lineage-specific differentiation, including the StemSpan CD34+ Expansion Supplement (10X) at catalogue #02691 and the StemSpan Myeloid Expansion Supplement (100X) at catalogue #02693. Added cytokines and cells should not exceed approximately 10% of the total culture volume. The bottle should be stored refrigerated, protected from light, and handled under aseptic technique.
