SOLIScript®_ Fast 1-step RT-qPCR Mix with UNG

A 4x concentrated one-step RT-qPCR mix from Solis BioDyne enables sensitive and accurate quantification of RNA targets by real-time RT-PCR using dual-labeled hydrolysis probes, and supports detection and quantitation of up to five targets simultaneously. The mix is supplied in a single tube, with nuclease-free water provided separately, and contains all components needed except RNA template, primers and probes.

It comprises SOLIScript® Reverse Transcriptase (active at temperatures up to 60°C), RiboGrip™ RNase Inhibitor (which inactivates RNase A, B and C(III)), HOT SolisFAST® DNA Polymerase with chemical hot-start and approximately 2-4 times faster extension rates than Taq DNA polymerase, plus heat-inactivated Salini UNG™ Uracil-N-Glycosylase together with dUTPs to prevent carry-over contamination. UNG is fully active between 25°C and 40°C and inactive at reaction temperatures above 50°C. The buffer provides 22.0 mM MgCl2 (5.5 mM MgCl2 in 1x solution).

Inhibitor tolerance and fast cycling enable flexible experiment design, and the mix supports co-detection of RNA viruses, DNA viruses and bacterial targets from one reaction. Manufacturer-reported multiplex RT-qPCR efficiencies range from 95% to 101% with R2>0.99. The product is compatible with most qPCR cyclers (a ROX version is available on request) and is supplied for research use only.
