Description

This reagent kit from Solis BioDyne is optimized for probe-based one-step RT real-time quantitative PCR assays. It contains all the components necessary — except template, primers, and probes — to perform cDNA synthesis and probe-based qPCR in a single tube. The kit is suitable for the detection and quantification of RNA targets with qPCR, for DNA/LNA hydrolysis probe-based assays, and for singleplex or duplex reactions.

The kit is supplied as three tubes: a 5x One-step Probe Mix containing HOT FIREPol® DNA Polymerase, reaction buffer, dNTPs (including dUTP), 15 mM MgCl₂, and an internal reference dye based on ROX; a 40x One-step SOLIScript® Mix containing SOLIScript® Reverse Transcriptase and RiboGrip® RNase Inhibitor; and nuclease-free water. The included SOLIScript® Reverse Transcriptase is an in silico-engineered thermostable enzyme active at higher temperatures (up to 60 °C), supporting highly specific results and work with templates that have high secondary structure. HOT FIREPol® DNA Polymerase is activated by a 10-minute incubation step at 95 °C, helping to prevent extension of non-specifically annealed primers and primer-dimers formed at low temperatures during qPCR setup. The kit is optimized for DNA/LNA hydrolysis probes based on the 5' flap endonuclease activity.

The reference dye is based on ROX, and the kit is compatible with most real-time qPCR cyclers, including both ROX-dependent and ROX-independent instruments, as well as those that do not require an internal reference dye for normalization of fluorescent signal. Routine storage is at −20 °C, and shipping, as well as temporary storage for up to one month at room temperature, has no detrimental effect on kit quality. The product is supplied for research use only.
