Hands-On hPSC Training in California

Delivered by STEMCELL Technologies as a 2-day intensive workshop, this winter-session course held in California trains researchers in the reprogramming and maintenance of human pluripotent stem cells (hPSCs). Sessions mix live bench demonstrations with supervised practical work on the feeder-free mTeSR™1 culture system, using non-integrating, self-replicating RNA reprogramming vectors to derive iPS cells from human fibroblasts.

Participants rotate through enzymatic passaging with Dispase, enzyme-free passaging with GCDR or ReLeSR™, and mechanical aggregate passaging targeting 50–200 µm clumps, then evaluate colony morphology and critical quality attributes such as pluripotency marker expression. Cryopreservation and thawing protocols are practiced alongside workflows for monitoring karyotypic drift, with troubleshooting focused on spontaneous differentiation, recovery of newly derived lines, and seeding-density optimization for routine confluence at day 5 to day 7.

Instruction is led by STEMCELL Technologies scientific faculty in small groups, with live demonstrations contrasting feeder-free and feeder-dependent culture systems. Each day concludes at approximately 4:30 pm. Attendees who complete the workshop receive a certificate of completion together with course materials and protocol references, making the program relevant for graduate students, postdoctoral researchers, and core facility staff working in regenerative medicine, developmental biology, cell therapy development, and related in vitro research applications.
