Description

An enzyme-free, cGMP reagent from STEMCELL Technologies intended for the dissociation and passaging of human pluripotent stem cells (hPSCs) as aggregates, without manual selection or scraping. ReLeSR™ selectively detaches undifferentiated cells from pluripotent stem cell cultures, leaving differentiated cells attached to the cultureware. Following incubation and gentle shaking or tapping, optimally-sized aggregates are generated for replating. The reagent can also rescue highly differentiated cultures; passaging with this reagent has been shown to convert a starting culture of ~50% undifferentiated cells into one containing >90% undifferentiated cells by the end of the next passage. It can additionally be used to isolate putative iPS cell colonies by selectively detaching undifferentiated cells while leaving non-reprogrammed cells behind.

The product is supplied as a clear, red liquid and is not classified as a hazardous substance or mixture according to the Globally Harmonized System (GHS) or the 2012 OSHA Hazard Communication Standard. It is intended for laboratory reagent use and is supplied by STEMCELL Technologies Canada Inc. (Vancouver, British Columbia, Canada). Storage conditions should follow the Product Information Sheet.

Workflow notes: A passaging incubation of approximately 4 minutes at 37°C is described for use with 6-well plates, yielding an optimal aggregate size of 50-200 µm. The surface area of 4 x 6 well plates (230 cm²) is comparable to that of a T225 flask (225 cm²).
