STEMCELL Technologies offers ReLeSR™, an enzyme-free, cGMP reagent developed for the dissociation and passaging of human pluripotent stem cells (hPSCs) as aggregates, without manual selection or scraping. By eliminating difficult and time-consuming steps, the reagent enables easy culture scale-up.

ReLeSR™ selectively detaches undifferentiated cells from pluripotent stem cell cultures, while differentiated cells remain attached to the cultureware. Optimally-sized aggregates are generated after shaking or tapping the cultureware. This selectivity also allows poor quality hPSC cultures containing ~50% undifferentiated cells to be rescued, with >90% undifferentiated cells present at the end of the next passage. Newly generated human iPS cell colonies can also be isolated by selectively detaching undifferentiated cells and leaving non-reprogrammed cells behind.

The reagent is intended for use as part of workflows involving frozen human pluripotent stem cell lines and cGMP, feeder-free maintenance media for human ES and iPS cells.
