QuantiNova SYBR Green RT-PCR Kit (500)

From QIAGEN, this one-step SYBR Green RT-PCR kit combines reverse transcription and quantitative PCR in a single-tube format for real-time analysis of RNA targets. The chemistry is suitable for total RNA from eukaryotes and prokaryotes, poly(A)- RNA, and in vitro-transcribed RNA, supporting applications such as expression profiling and routine RT-qPCR work.

The 2x QuantiNova SYBR Green RT-PCR Master Mix contains QuantiNova DNA Polymerase (a modified recombinant 94 kDa enzyme originally isolated from Thermus aquaticus), QuantiNova SYBR Green RT-PCR Buffer, and SYBR Green I. The buffer provides Tris-HCl, KCl, NH4SO4, and MgCl2 together with the Q-Bond additive, which enables short cycling steps without loss of PCR sensitivity. SYBR Green I binds double-stranded DNA, with excitation and emission maxima at 494 nm and 521 nm respectively, and is compatible with any real-time cycler. The 100x QuantiNova RT Mix provides HotStaRT-Script Reverse Transcriptase — a modified recombinant 77 kDa reverse transcriptase paired with an RT-blocker and an RNase Inhibitor — so that cDNA synthesis begins at the 50°C RT step while the DNA Polymerase is kept inactive until activation within 2 minutes at 95°C.

Reaction setup can be performed at room temperature, and the assembled reaction can be held at room temperature for up to 2 hours without performance loss. The kit also includes QuantiNova Internal Control RNA for monitoring successful reverse transcription, QuantiNova Yellow Template Dilution Buffer, QuantiNova ROX Reference Dye (for Applied Biosystems instruments requiring a passive reference), and RNase-Free Water. Kits are shipped on dry ice and stored at –15 to –30°C protected from light; the Master Mix, Yellow Template Dilution Buffer, and ROX Reference Dye may alternatively be stored at 2–8°C for up to 12 months.
