Airway epithelial cell expansion is supported through a defined, serum-free, animal component-free culture medium from STEMCELL Technologies, formulated for the sustained proliferation of primary human bronchial, nasal, and small airway epithelial cells. PneumaCult-Ex Medium preserves the basal cell compartment that underpins regenerative capacity in the respiratory mucosa, allowing researchers to maintain passage-to-passage consistency without animal serum variability.

Researchers apply this medium in both 2D monolayer and 3D organoid formats for respiratory disease modeling, including chronic obstructive pulmonary disease, cystic fibrosis, asthma, and SARS-CoV-2 entry studies. Paired with PneumaCult-ALI Medium, expanded cells differentiate at an air-liquid interface into pseudostratified mucociliary epithelium containing ciliated cells, goblet cells, and basal cells, replicating in vivo airway architecture for translational investigation.

Supplied as a 450 mL basal medium bottle accompanied by a 50 mL supplement vial, the working solution is prepared by combining the supplement with basal medium at a 1:9 ratio, and the complete medium should be stored refrigerated and protected from light to maintain growth factor activity. Prepared working solution remains stable for up to two weeks under cold storage, and 1 mL aliquots are suitable for downstream applications such as in vitro drug toxicity screening, respiratory toxicology assays, gene editing workflows, and disease modeling.

Each lot is performance-tested on primary human bronchial epithelial cells to confirm consistent expansion across multiple passages, supporting reproducible protocols for lung research, smoke and vaping exposure studies, and airway inflammation investigations.
