Description

From STEMCELL Technologies, PneumaCult™ Apical-Out Airway Organoid (AOAO) Medium is a serum- and BPE-free medium for the differentiation of human primary bronchial epithelial cells (HBECs) or human airway epithelial cells (HAECs) to mature apical-out airway organoids in 15 days. When fully differentiated, the organoids exhibit outward-facing ciliated cells characteristic of the apical side of the epithelium lining the airways in vivo.

This kit version pairs the AOAO Medium with AggreWell™400 24-well plates to support aggregate formation during the workflow. Culture of HBECs or HAECs in PneumaCult™-Ex Plus Medium prior to inducing organoid differentiation with PneumaCult™ AOAO Medium constitutes a complete serum- and BPE-free workflow. Applications of apical-out airway organoids include infectious disease modeling and high-throughput drug screening in in vitro, three-dimensional (3D) model systems.

The kit is supplied as three components sold together: PneumaCult™ Apical-Out Airway Organoid Basal Medium (450 mL, stored at 2 - 8 °C, stable for 12 months from date of manufacture), 10X Supplement (50 mL, stored at -20 °C, stable for 2 years from date of manufacture), and 1000X Supplement (500 µL, stored at -20 °C, stable for 12 months from date of manufacture). The 10X Supplement contains material derived from human plasma; donors have been tested and found negative for HIV-1 and -2, hepatitis B, and hepatitis C prior to donation, but the product should be considered potentially infectious and handled with universal precautions. The 1000X Supplement is light sensitive and is dissolved in dimethyl sulfoxide (DMSO), a strong solvent and skin penetrant that can transport many substances through the skin and penetrate some protective glove materials including latex and silicone.
