PneumaCult Alveolar Organoid Diff Medium

The PneumaCult™ Alveolar Organoid Differentiation Medium from STEMCELL Technologies is a two-component, serum-free cell culture medium used in the second stage of the PneumaCult™ Alveolar Organoid Media workflow to differentiate human alveolar epithelial type II (ATII) organoids into alveolar epithelial type I (ATI) organoids for downstream applications. Following expansion of ATII organoids in PneumaCult™ Alveolar Organoid Expansion (AvOE) Medium, cultures are transferred to the differentiation medium for 10 additional days to generate ATI organoids, reportedly yielding over 85% purity.

Differentiated cells show decreased ATII marker expression alongside strong up-regulation of ATI markers including RAGE/AGER, HT1-56, and GPRC5a, as demonstrated by flow cytometry and immunohistochemistry. The medium is supplied as PneumaCult™ Alveolar Organoid Differentiation Basal Medium (90 mL) plus PneumaCult™ Alveolar Organoid Differentiation 10X Supplement (10 mL), a format designed to maximize experimental reproducibility. It is compatible with primary isolated fresh or cryopreserved ATII cells, as well as high-quality commercially available alveolar epithelial cell sources. Differentiated ATI organoids and derived monolayers are suited to alveolar biology research, infectious disease studies, and drug screening, and the cultures express SARS-CoV-2 entry-associated proteins TMPRSS2 and ACE2.
