PneumaCult Airway Organoid Seeding Supp

A serum-free seeding supplement from STEMCELL Technologies formulated to support initial establishment and clonal expansion of primary human airway basal epithelial cells embedded in Matrigel dome cultures. Applied during the first three to seven days of airway organoid formation, it sustains proliferation of p63-positive, KRT5-positive basal cells before they transition to the maturation phase driven by PneumaCult Airway Organoid Growth Supplement.

Supplied as a single-use vial, the supplement is aliquoted upon arrival and stored frozen at -20 degrees C to safeguard activity through the seeding workflow. Each aliquot is thawed fresh per experiment, and repeated freeze-thaw cycles are avoided to preserve consistent organoid-initiating capacity. Intended for in vitro research use only, this reagent is widely applied across respiratory biology, including SARS-CoV-2 airway infection modeling, cystic fibrosis, COPD, idiopathic pulmonary fibrosis, and characterization of basal airway stem cells. It is combined with PneumaCult Airway Organoid Basal Medium during the seeding stage and removed when cultures are switched to the growth medium formulation for long-term passaging and downstream differentiation assays.
