Description

A serum-free organoid growth medium system from STEMCELL Technologies, designed for the establishment, expansion, and long-term maintenance of human pancreatic duct organoids. The kit is supplied as a complete culture medium package containing PancreaCult Organoid Basal Medium (Human) and PancreaCult Organoid Growth Supplement (Human), and is intended for use following initiation in PancreaCult Organoid Initiation Medium (OIM), or alternatively for a completely serum-free workflow initiated directly in OGM.

Organoids are established from fresh or cryopreserved islet-depleted exocrine fractions, or from dissociated tumor tissue. Once established, cultures can be passaged, cryopreserved for future experiments, and adapted to a range of formats including two-dimensional Transwell monolayer cultures, dilute Corning Matrigel suspension cultures, and high-throughput assay-compatible culture formats. The growth medium also allows for the transient removal of epidermal growth factor (EGF) to support the depletion of normal cells from K-Ras-activated pancreatic tumor tissue cultures. Established organoids from normal tissue display a cystic morphology with a hollow lumen and apical-basolateral orientation, and maintain expression of several duct cell markers.

Applications of the system include studying the development and function of duct epithelium, modeling pancreatic diseases such as cancer or cystic fibrosis, and performing targeted drug screens. The Basal Medium is stored at 2 - 8°C with a shelf life of 2 years from the date of manufacture, while the Growth Supplement is stored at -20°C with a shelf life of 18 months from the date of manufacture. The product was developed under a licence to intellectual property owned by Hubrecht Organoid Technology (HUB) and is sold for research use only.
