Description

This small molecule from STEMCELL Technologies is a selective and potent, ATP-competitive inhibitor of the fibroblast growth factor receptor (FGFR). It acts on both FGFR3 and FGFR1 with IC₅₀ values of 5 nM and 21.5 nM respectively, and also inhibits FGFR2, FGFR4, and vascular endothelial growth factor receptor 2 (VEGFR2). The compound is approximately 1000 times more potent than another common FGFR inhibitor, SU5402, while showing little to no activity against PDGFR, EGFR, MEK, or PKC.

Supplied as a crystalline solid with a purity of ≥ 98%, the product has a molecular weight of 523.7 g/mol and the chemical name N-[2-[[4-(diethylamino)butyl]amino]-6-(3,5-dimethoxyphenyl)pyrido[2,3-d]pyrimidin-7-yl]-N'-(1,1-dimethylethyl)-urea. The CAS number is 219580-11-7. The compound is stable at -20°C as supplied, should be protected from prolonged exposure to light, and remains stable for 12 months from the date of receipt. It has low solubility in aqueous media, with solubility in DMSO of ≤ 20 mM. For use as a cell culture supplement, the stock solution should be diluted into culture medium immediately before use, with final DMSO concentration above 0.1% avoided due to potential cell toxicity.

In published research, the inhibitor has been used to suppress the differentiation of mouse ES cells and maintain the undifferentiated state, to prevent excision-mediated differentiation of mouse induced pluripotent stem cells generated using piggyBac transposons, and to promote reprogramming of human embryonic stem cells to a naïve state in combination with other factors. It also blocks neural differentiation of mouse ES cells and promotes differentiation of human ES cells, though not when they are in a naïve or ground state. The product is hazardous and is intended for research use only.
