From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit is intended for the in vitro quantitative determination of mouse Neuregulin 4 (NRG4) in serum, plasma, tissue homogenates or other biological fluids.

The assay uses the sandwich enzyme immunoassay technique. An antibody specific for NRG4 is pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, where any NRG4 present is captured by the immobilised antibody. After incubation and washing, a Biotinylated Anti-NRG4 Antibody is added and binds the captured NRG4. HRP-Avidin conjugate is then introduced, followed by further incubation and washing. A TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of NRG4 captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader against a standard curve.

Reported sensitivity is <0.128 ng/ml and the assay range is 0.312–20 ng/ml. The kit is read on a colorimetric microplate reader, runs on a pre-coated microplate of 12 x 8 well strips, and has a total assay time of 4h 30m. Kit components should be stored at +4°C or –20°C as instructed, and the product must not be used past its expiration date. In validation studies (n=5), average recovery was 91% in serum (range 80–102%), 90% in EDTA plasma (range 81–100%) and 84% in heparin plasma (range 80–89%). Dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions across serum, EDTA plasma and heparin plasma.
