Annexin V-FITC Apoptosis Detection Kit

The Annexin V-FITC Apoptosis Detection Kit from Antibodies.com enables sensitive identification of apoptotic cells by exploiting the early translocation of phosphatidylserine to the outer leaflet of the plasma membrane. The FITC-conjugated Annexin V reagent binds phosphatidylserine in a calcium-dependent manner and is read in the FITC channel of a flow cytometer (488 nm excitation, emission collected around 530 nm with a 30 nm bandpass filter), making the kit well suited to flow cytometry and fluorescence microscopy workflows.

For each assay, 100 uL of cell suspension at 1 x 10^6 cells/mL is incubated with 5 uL Annexin V-FITC in the supplied calcium-containing binding buffer for 10-15 minutes at room temperature in the dark. A propidium iodide counterstain is typically added immediately before acquisition to discriminate early apoptotic (Annexin V-positive, PI-negative) from late apoptotic and necrotic (Annexin V-positive, PI-positive) populations. Each pack supplies sufficient reagent for 100 tests and includes binding buffer; store refrigerated and protect from light, and avoid freeze-thaw cycles of the conjugated reagent to preserve fluorescence intensity.
