The Antibodies.com Annexin V-PE Apoptosis Detection Kit is a flow cytometry reagent set for identifying apoptotic cells through the externalization of phosphatidylserine (PS). The kit uses recombinant Annexin V conjugated to R-Phycoerythrin (PE), a ~35-36 kDa calcium-dependent phospholipid-binding protein, which labels PS exposed on the outer leaflet of the plasma membrane during early apoptosis. Combined with propidium iodide counterstaining, researchers can discriminate viable (Annexin V-/PI-), early apoptotic (Annexin V+/PI-), and late apoptotic or necrotic (Annexin V+/PI+) populations on FL1 (PE) and FL2 (PI) channels.
The 100-test configuration is supplied with the Annexin V-PE conjugate vial, propidium iodide solution, and 10X binding buffer; tubes should be handled with a micropipette in a biosafety hood and stained cell suspensions analyzed within 1 hour. Each component should be stored at 4 °C, protected from light, and equilibrated to room temperature before use. Intended for in vitro research use only, the protocol is compatible with suspension cell lines, freshly isolated primary cultures, and thawed cryopreserved samples prepared by standard washing and resuspension steps.
