This hands-on training course teaches researchers the complete workflow for establishing and maintaining mouse intestinal organoid cultures. Delivered over 2 days in a laboratory setting, the program covers crypt isolation from murine small intestine, embedding in Matrigel domes, and feeding with IntestiCult medium for sustained 3D growth.
Participants receive step-by-step instruction on passaging, cryopreservation, and downstream applications including gene editing and disease modeling. Each attendee completes practical bench work using microscopes, biosafety cabinets, and freshly isolated tissue, with a protocol handbook and a starter kit of reagents provided.
The course targets graduate students, postdocs, and core facility staff seeking reproducible organoid culture skills. STEMCELL Technologies instructors guide small groups through troubleshooting, with emphasis on contamination prevention, dome morphology assessment, and scaling cultures for intestinal research. Lunch and refreshments are included on both training days.
