Mouse Catechol-O-Methyltransferase ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay is intended for the in vitro quantitative determination of mouse COMT (Catechol-O-Methyltransferase) in tissue homogenates, cell lysates, or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) and employs the sandwich enzyme immunoassay technique, in which an antibody specific for COMT captures the analyte, followed by a Biotinylated Anti-COMT Antibody and an HRP-Avidin conjugate. A TMB substrate produces a colour reaction that is read at 450 nm on a colorimetric microplate reader.

The assay has a reported sensitivity of <0.055 ng/ml and an assay range of 0.156–10 ng/ml, with a total assay time of 4 h 30 m. Reported mean recovery values are 91% for serum (n=5, range 80%–102%), 90% for EDTA plasma (n=5, range 81%–100%) and 84% for heparin plasma (n=5, range 80%–89%). Linearity across dilutions of 1:2, 1:4, 1:8 and 1:16 gave ranges of 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5).

Kit components include a pre-coated 96-well microplate, lyophilised standard, Detection Solution A and B, 30× wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components should be stored as instructed at +4°C or -20°C and the kit must not be used past its expiration date.
