A serum-free methylcellulose-based medium from STEMCELL Technologies supports colony-forming unit (CFU) assays of human hematopoietic progenitor cells, enabling growth and enumeration of erythroid and myeloid lineage colonies — including BFU-E, CFU-E, CFU-GM, and CFU-GEMM — in defined, xeno-free culture conditions.

The ready-to-use 100 mL formulation is supplemented with recombinant human cytokines (rh SCF, rh IL-3, rh GM-CSF, and erythropoietin) along with BSA, human insulin, human transferrin, and other defined supplements. No additional supplementation is required: cells resuspended in Iscove's Modified Dulbecco's Medium are added directly and plated into standard 35 mm culture dishes for incubation at 37 °C in a 5% CO₂ humidified atmosphere.

Typical applications include hematopoietic toxicity screening, characterization of cord blood, bone marrow, mobilized peripheral blood, or purified CD34+ cell populations, and studies of progenitor expansion, differentiation, or lineage commitment. Erythroid and myeloid colonies are scored after approximately 14 days of culture using an inverted microscope.

Store frozen at -20 °C; once thawed, aliquot into single-use working volumes and avoid repeated freeze-thaw cycles to preserve recombinant cytokine activity. Each 100 mL bottle provides sufficient medium for roughly 50 to 60 standard 35 mm dishes plated at 1.1 mL per dish, making it a practical format for routine research use only workflows in academic and translational hematology laboratories.
