MethoCult SF H4436, 100mL

Serum-free methylcellulose medium supplied by STEMCELL Technologies in a 100 mL bottle supports human hematopoietic colony-forming unit assays. The MethoCult SF formulation is engineered for in vitro culture of erythroid, granulocyte/macrophage, and multilineage progenitor colonies (BFU-E, CFU-GM, CFU-GEMM) from bone marrow, peripheral blood, or cord blood mononuclear cells within a single culture. Recombinant human cytokines—SCF, IL-3, IL-6, G-CSF, GM-CSF, and erythropoietin—are pre-titrated to remove the lot-to-lot variability of serum-supplemented systems. Cells are typically plated at 2 to 3 mL per 35 mm dish and incubated at 37 °C in a humidified 5% CO2 atmosphere; colonies are scored on day 14 with an inverted microscope. Applications include drug screening, genotoxicity testing, characterization of normal versus malignant hematopoiesis, and validation of gene-editing protocols in hematopoietic stem and progenitor cells.
