A serum-free, methylcellulose-based colony-forming unit (CFU) assay medium from STEMCELL Technologies supplies recombinant human cytokines including stem cell factor (SCF), interleukin-3 (IL-3), interleukin-6 (IL-6), granulocyte colony-stimulating factor (G-CSF), and granulocyte-macrophage colony-stimulating factor (GM-CSF). The formulation does not contain erythropoietin (EPO), making it appropriate for assays that evaluate cytokine-driven myeloid and erythroid progenitor responses independent of exogenous EPO.

MethoCult SF is prepared for use with mononuclear cells isolated from bone marrow, peripheral blood, or cord blood, and supports the growth of erythroid burst-forming units (BFU-E), granulocyte-macrophage colony-forming units (CFU-GM), and multipotent granulocyte/erythrocyte/macrophage/megakaryocyte colonies (CFU-GEMM). The 80 mL bottle provides sufficient volume for multiple standard cultures; one bottle supports approximately 72 assays when dispensed at 1 mL per well in a 6-well plate, or roughly 50 assays at 1.1 mL per well.

For handling, the product should be stored frozen at -20 °C and protected from light. Repeated freeze-thaw cycles should be avoided; once thawed, aliquots should be used promptly. Each bottle is supplied as a single-use liquid formulation ready to be combined with cells and dispensed into culture dishes for hematopoietic colony enumeration in research use only applications.
