Methylcellulose-based colony assay medium from STEMCELL Technologies supports the growth and enumeration of mouse hematopoietic progenitor cells in semi-solid culture. Formulated without erythropoietin (EPO), this 90 mL bottle is intended for assaying non-erythroid colony-forming units such as CFU-G, CFU-M, and CFU-GM from mouse bone marrow, spleen, or fetal liver cell suspensions. Researchers typically plate cells in 35 mm dishes or 6-well plates, incubate at 37 °C with 5% CO₂, and score colonies after 7–14 days using an inverted microscope. The medium is supplied frozen and should be stored at -20 °C; thaw at room temperature or briefly in a 37 °C water bath before use and avoid repeated freeze-thaw cycles. Each bottle provides sufficient volume for multiple 35 mm assay dishes at standard plating densities. Colonies can be further characterized by Wright-Giemsa staining of individually picked clusters to confirm granulocyte or macrophage lineage identity. For research use only.
