MethoCult H4435 Enriched, 3mL

A viscous, methylcellulose-based hematopoietic colony assay medium supplied as 3 mL single-use vials by STEMCELL Technologies. Each vial supports simultaneous growth of erythroid BFU-E, granulocyte-macrophage CFU-GM, and multilineage CFU-GEMM colonies from human bone marrow, mobilized peripheral blood, or cord blood mononuclear cells. The pre-mixed formulation contains recombinant human SCF, GM-CSF, G-CSF, IL-3, IL-6, erythropoietin, and Flt-3 Ligand at optimized working concentrations, so no separate cytokine additions are required before plating.

For the assay, mononuclear cells are suspended directly into the vial contents and dispensed at 1 mL per 35 mm dish, then incubated at 37°C in humidified 5% CO₂ for 14 to 16 days. Colonies are then scored under an inverted microscope without further staining. Recommended plating density is roughly 2 × 10⁵ mononuclear cells per dish. Vials ship frozen and should be stored at -20°C; once thawed, the medium remains stable for up to one month when refrigerated.
