MethoCult H4034 Optimum, 24x3mL/tube

A methylcellulose-based semi-solid medium from STEMCELL Technologies supplied as 24 pre-filled 3 mL tubes, ready for direct plating in colony-forming unit (CFU) assays of human hematopoietic progenitor cells. The MethoCult H4034 Optimum recipe combines a viscous methylcellulose matrix with a pre-titrated cytokine cocktail, removing the requirement for users to supplement with recombinant human SCF, FLT3 ligand, IL-3, IL-6, and erythropoietin prior to plating.

Each tube serves as a single-use seed vessel for mononuclear cells isolated from bone marrow aspirate, mobilized peripheral blood, or umbilical cord blood. After plating into 35 mm culture dishes and incubating at 37°C in 5% CO2 for 10 to 14 days, primitive erythroid (BFU-E), granulocyte-macrophage (CFU-GM), and multilineage (CFU-GEMM) colonies can be counted under a standard inverted microscope.

Tubes arrive frozen and should be stored at -20°C, protected from light, then thawed overnight at refrigerator temperature or briefly at room temperature before use. The pre-portioned single-use format minimizes hands-on preparation time and inter-lot variability compared with manually supplemented methylcellulose systems, supporting reproducible in vitro research applications in hematopoiesis, drug screening, and cell therapy development.
