A methylcellulose-based colony assay medium supplied in a 3 mL single use vial, this research reagent from STEMCELL Technologies enables enumeration of human hematopoietic progenitor cells across erythroid, myeloid, and multilineage lineages. The pre-formulated matrix contains recombinant cytokines that support growth of BFU-E, CFU-E, CFU-G, CFU-M, CFU-GM, and CFU-GEMM colonies derived from human bone marrow, cord blood, or mobilized peripheral blood mononuclear cells without the need for a separate feeder layer.

Store vials at -20 degrees C or colder to preserve cytokine activity; thaw immediately before use and avoid repeated freeze-thaw cycles. Standard plating involves combining 1.1 mL of the methylcellulose mixture with a defined cell suspension in 35 mm culture dishes, which are incubated at 37 degrees C in a humidified 5% CO2 incubator for 14 to 16 days before colony scoring under an inverted microscope.
