A methylcellulose-based medium formulated by STEMCELL Technologies to support optimal growth of human granulocyte-macrophage progenitor cells (CFU-GM, CFU-M, CFU-G) in colony-forming unit (CFU) assays. It is intended for detecting and quantifying human hematopoietic progenitor cells in bone marrow, mobilized peripheral blood, peripheral blood, and cord blood samples, and is recommended for CD34+ enriched cells, mononuclear cells, and cells prepared by other purification methods.

Formulation components include Iscove's MDM, methylcellulose, fetal bovine serum, bovine serum albumin, 2-mercaptoethanol, recombinant human stem cell factor, rh GM-CSF, rh G-CSF, and rh Interleukin 3. Each batch is manufactured using aseptic technique, sterility tested according to USP methods, and quality-control performance tested in CFU assays using human bone marrow, cord blood, or peripheral blood samples. A Certificate of Analysis is available upon request.

Intended for in vitro diagnostic use by trained professionals observing good laboratory practices. Store at -15 to -25°C; the product remains stable at this range until the expiry date on the label and should not be repeatedly freeze-thawed. For assay setup, a 10X concentrated cell suspension is added to the medium at a 1:10 v/v ratio, dispensed into culture dishes, and incubated at 37°C with 5% CO2 and ≥95% humidity for 14-16 days, after which colonies are counted using an inverted microscope.
