A methylcellulose-based medium with recombinant cytokines from STEMCELL Technologies, intended for use in colony-forming unit (CFU) assays to detect and quantify human hematopoietic progenitor cells in bone marrow, mobilized peripheral blood, peripheral blood, and cord blood samples. It is recommended for CD34+ enriched cells, mononuclear cells, and cells isolated by other purification methods.

The formulation has been designed to support optimal growth of erythroid progenitor cells (CFU-E and BFU-E); granulocyte-macrophage progenitor cells (CFU-GM, CFU-M, CFU-G); and multi-potential granulocyte, erythroid, macrophage, megakaryocyte progenitor cells (CFU-GEMM). Components include Iscove's MDM, methylcellulose, fetal bovine serum, bovine serum albumin, recombinant human stem cell factor, recombinant human GM-CSF, recombinant human G-CSF, recombinant human Interleukin 3, recombinant human Interleukin 6, and recombinant human erythropoietin (EPO).

The medium is manufactured using aseptic technique, sterility tested according to USP methods, and quality control performance tested in CFU assays using human bone marrow, cord blood, or peripheral blood samples. It should be stored at -15 to -25°C and is intended for in vitro diagnostic use by trained personnel observing good laboratory practices. The product contains material of animal origin and may be a potential irritant to eyes, respiratory system, and skin; it may also be harmful if ingested and may cause allergic reaction in sensitized individuals.
