MethoCult Cell Wash Medium,100mL

From STEMCELL Technologies, this cell wash medium is formulated for use as a wash and dilution reagent in colony-forming unit (CFU) assays of human hematopoietic progenitor cells, in combination with MethoCult™ Express methylcellulose medium. It is intended for in vitro diagnostic use and carries a CE IVD marking.

The medium is recommended for washing and diluting cell samples prior to CFU assay set-up with human cord blood (CB) sources, including red blood cell-depleted CB samples, whole CB samples that have been cryopreserved and thawed, and CB mononuclear cells. A 10X concentrated cell suspension is typically prepared in the wash medium before cells are combined with MethoCult™ at a 1:10 v/v ratio to ensure correct medium viscosity. For example, a sample of 5 x 10⁵ cells/mL in the wash medium corresponds to a plating concentration of 5 x 10⁴ cells per dish. Plating cells at 2-4 different densities is recommended to yield approximately 25-120 colonies per 35 mm dish. Colonies containing at least 20 cells can be counted after 7 days of culture; if cultures are maintained for 14-16 days, erythroid (BFU-E), granulocyte/macrophage (CFU-GM, CFU-G, CFU-M) and multi-potential (CFU-GEMM) progenitors can be distinguished.

Once human cells are combined with the product, the mixture should be treated as potentially biohazardous, and the medium should be handled by trained personnel observing good laboratory practices.
