Description

STEMCELL Technologies has developed a complete cell culture medium — IntestiCult™-SF Organoid Growth Medium (Human) — designed to support the establishment, expansion, and long-term maintenance of human intestinal organoids. The formulation is serum-free and conditioned medium-free, providing a defined culture environment for organoids derived from human intestinal crypts.

Isolated intestinal crypts rapidly form complex organoids that incorporate a functional lumen enclosed by a polarized epithelial cell layer containing known cell types of the adult intestinal epithelium. Maturing organoids establish protruding buds resembling intestinal crypts of in vivo tissues. The medium supports proliferation of intestinal epithelial stem cells from patient biopsies and enables expansion of cultures derived from fresh tissue or from already established frozen stocks. The manufacturer reports that cultures grown in this medium display morphology and genetic expression profiles comparable to those grown in the brand's standard Organoid Growth Medium (Human), with more efficient expansion during extended culture.

Applications include studying the development and function of the intestinal epithelium, modelling intestinal diseases, performing targeted molecule screens, and investigating adult stem cell properties and regenerative therapy approaches. Organoids can be further differentiated using the brand's Organoid Differentiation Medium (Human). The basal medium incorporates DMEM/F-12 with 15 mM HEPES buffer. Store at -20°C; the product is stable for 2 years from the date of manufacture on the label. The product contains material derived from human plasma, with donors tested and found negative for hepatitis B surface antigen (HBsAg), HIV-1 antibodies, and/or HIV-1 antigen.
