STEMCELL Technologies IntestiCult OGM Mouse Kit delivers a defined, serum-free culture system for establishing and long-term expansion of murine intestinal organoids from isolated crypts. Each complete kit contains a 90 mL bottle of basal medium together with two 5 mL vials of frozen growth factor supplements, providing every component needed to support Lgr5-positive stem cell populations embedded in a dome matrix. The serum-free formulation enables reproducible passaging every 7 to 10 days, with a recommended split ratio of 1:6 for small intestinal cultures and 1:2 for colonic organoids, which are typically passaged once cultures reach approximately 150 organoids per well. Researchers apply the medium across mucosal immunology studies, host-microbe interaction assays, CRISPR-mediated gene editing workflows, drug screening campaigns, and disease modeling relevant to epithelial biology. The kit integrates with downstream histology, qPCR, whole-mount imaging, and single-cell sequencing readouts, supporting both basic discovery and translational research programs. Basal medium should be stored refrigerated; supplement vials must remain frozen at -20 °C, protected from light, with repeated freeze-thaw cycles avoided to preserve growth factor activity. Crypts harvested from the small intestine generally initiate budding within 2 to 4 days in culture, whereas colonic preparations begin forming organoids within 7 to 10 days. The system is intended for in vitro research use only and the supplied components contain no antibiotics.
