Description

STEMCELL Technologies manufactures this liquid cell culture medium designed for the culture of Wnt-independent cancer-derived intestinal organoids, including colorectal cancer-derived organoids, with the protocol developed in collaboration with Hubrecht Organoid Technology. The product is described in its safety documentation as a laboratory reagent intended for establishing and maintaining 3D organoid models from tumor biopsy samples.

According to the manufacturer's safety data sheet, the medium is a clear, red-orange liquid for which no odor data are available. It is not classified as a hazardous substance or mixture according to the Globally Harmonized System (GHS) and is stable under recommended transport and storage conditions. The product contains material derived from human plasma; donors have been tested and found negative for HIV-1 and -2, hepatitis B, and hepatitis C prior to donation, although the medium should still be considered potentially infectious and handled using biohazard-appropriate controls.

For storage, the basal medium may be used immediately or aliquoted and stored at -20°C for up to 3 months; once thawed, aliquots should be used immediately and not re-frozen. If combined with DMEM/F12 with 15 mM HEPES and not used immediately, the prepared medium can be stored at 2 - 8°C for up to 1 week. During culture, the growth medium should be replaced every 2 days, and cultures can be passaged every 6 - 12 days. Tumors that do not carry an activating mutation of the Wnt pathway cannot be maintained as organoids using this method.
