ImmunoCult-XF

A GMP-manufactured, serum-free and xeno-free cell culture medium from STEMCELL Technologies, ImmunoCult-XF is designed for the activation and expansion of human immune cells, including T cells, in research and clinical applications. The medium is produced under Good Manufacturing Practices with raw material traceability, environmental monitoring, aseptic process simulation, and validated test methods covering sterility, mycoplasma, and endotoxin testing; an FDA Master File is referenced, along with Certificates of Analysis and Origin available on request.

For T cell workflows, ImmunoCult-XF can be combined with the GMP ImmunoCult Human CD3/CD28 T Cell Activator or the GMP ImmunoCult Human CD3/CD28/CD2 T Cell Activator and supplemented with cytokines such as IL-2, or IL-7 with IL-15, to drive proliferation and upregulation of the activation marker CD25. Representative manufacturer protocols seed T cells at 1 x 10^6 cells/mL in ImmunoCult-XF supplemented with 180 IU/mL Human Recombinant IL-2 and stimulate with 25 µL/mL of the CD3/CD28/CD2 activator, harvesting at Day 10 or Day 12 depending on the culture format.

Reported performance figures include an average viability of 92.8 ± 1.8% and a total fold expansion of 1497 ± 143 by Day 12 under a low cell density static protocol (n = 11); 89.3 ± 2.3% viability and a total fold expansion of 67 ± 7 by Day 12 under a high cell density static protocol (n = 4); and 92.7 ± 0.7% viability with a total fold expansion of 503 ± 51 by Day 10 in the Xuri Cell Expansion System W25 (n = 4). The medium has also been used to support natural killer (NK) cell culture when combined with ImmunoCult NK Cell Expansion Supplement and ImmunoCult NK Cell Expansion Coating Material.
