ImmunoCult Ms Th2 Diff Suppl, 1mL

A differentiation supplement from STEMCELL Technologies formulated to promote the differentiation of mouse naïve CD4+ T cells into Th2 cells. The product combines recombinant mouse cytokines with a blocking monoclonal antibody, including recombinant mouse interleukin-2 (IL-2), recombinant mouse interleukin-4 (IL-4), and rat anti-mouse interferon-gamma (anti-IFN-γ).

The supplement is intended for use with RPMI 1640 medium containing fetal bovine serum and other additives, together with anti-mouse CD3 and anti-mouse CD28 monoclonal antibodies as activating agents. It is supplied as a 100X concentrate for addition to medium at a 1 in 100 dilution, and is optimized for polarization of naïve CD4+ T cells isolated from the spleen of BALB/c mice.

Store at -20°C; the product is stable for 18 months from the date of manufacture on the label. According to the manufacturer's data, naïve CD4+ T cells cultured with the supplement for 6 days produced a mean of 25 ± 3% CD4+IL-4+GATA3+ cells versus 4 ± 1% in non-polarized cultures (p < 0.001; n = 10), and a 62-fold expansion of total nucleated cells (n = 11, p < 0.001).
