ImmunoCult Ms Th1 Diff Suppl, 1mL

A differentiation supplement from STEMCELL Technologies formulated to promote the differentiation of mouse naïve CD4+ T cells into Th1 cells. The product contains a combination of recombinant mouse cytokines and a blocking monoclonal antibody, specifically recombinant mouse IL-2, recombinant mouse IL-12, and rat anti-mouse IL-4, and is supplied as a 100X concentrate intended for addition to RPMI 1640 medium containing fetal bovine serum together with anti-mouse CD3 and anti-mouse CD28 monoclonal antibodies as activating agents.

It is optimized for polarization of naïve CD4+ T cells isolated from the spleen of C57BL/6 mice. Under the recommended culture conditions, the mean proportion of CD4+IFNγ+T-Bet+ cells is 44 ± 6% compared with 2 ± 1% in non-polarized cultures (p < 0.001; n = 13), and polarized cultures show a 4.5-fold expansion of total nucleated cells (mean ± SEM; n = 11–20; *p < 0.05).

The supplement is a clear, colorless liquid that should be stored at -20°C and is stable for 18 months from the date of manufacture. It is intended for research use only.
