ImmunoCult Human Th1 Diff Supplement

STEMCELL Technologies offers a serum-free culture supplement designed to promote the robust activation, expansion, and differentiation of peripheral blood-derived, naïve, CD4+ human T cells into Th1 cells. The supplement contains a combination of human cytokines and a blocking antibody, including recombinant human interleukin 12 (IL-12), recombinant human interleukin 18 (IL-18), and a mouse anti-human interleukin 4 (anti-IL-4) reagent.

It is intended for use with ImmunoCult™-XF T Cell Expansion Medium and ImmunoCult™ Human CD3/CD28 T Cell Activator, and is supplied as a 50X concentrate that can be added directly to medium after thawing and mixing. According to the manufacturer, the supplement supports significantly higher levels of memory T cells and up to 1.6X higher expansion of IFN-γ-producing CD4+ T cells in comparison to a competitor Th1 differentiation kit. The product is formulated for the induction of human Th1 differentiation under defined conditions.

The supplement should be stored at -20°C and is stable for 12 months from the date of manufacture on the label. In the recommended protocol, purified human naïve CD4+ T cells are seeded at 1 x 10^6 cells/mL in complete Th1 Differentiation Medium, incubated at 37°C and 5% CO₂, and adjusted to 1 x 10^6 cells/mL every 3 - 4 days with fresh complete medium for 7 days, after which differentiated Th1 cells are ready for downstream applications.
