ImmunoCult HuCD3/CD28/CD2 TCellAct, 2mL

STEMCELL Technologies has developed this reagent for activating and expanding human T cells without the use of magnetic beads, feeder cells, or antigen. The product is composed of soluble antibody complexes that bind CD3, CD28, and CD2 cell surface ligands, and cross-linking of these ligands delivers the primary and co-stimulatory signals required for T cell activation. It is described as a highly stable, filter-sterilized soluble reagent that provides a gentle activation stimulus while maintaining high viability of activated and expanded T cells.

The reagent contains three monospecific antibody complexes: anti-human CD3, anti-human CD28, and anti-human CD2. It should be stored at 2 - 8°C and remains stable until the expiry date printed on the label.

In the manufacturer's protocol, viable human T cells are seeded at 1 x 10^6 cells/mL, and 25 µL/mL of the activator is added to the cell suspension, followed by incubation at 37°C and 5% CO2. According to the data provided, the frequency of CD25+ cells was 5.6 ± 2.4% on day 0 and rose to 88.8 ± 3.2% after 3 days of culture (mean ± SD). EasySep-isolated human T cells were expanded over 12 days in this system. Activated T cells can typically be used for gene editing 2 - 3 days after activation.
