ImmunoCult Hu CD3/CD28 TCell Act, 10 mL

STEMCELL Technologies offers this reagent as a cGMP human T cell activation and expansion reagent. It is designed to activate and expand human T cells using soluble antibody complexes, with the goal of maintaining high cell viability throughout the culture process.

In published experiments, human T cells isolated with the EasySep™ Human T Cell Isolation Kit were stimulated with this activator and cultured in ImmunoCult™-XF T Cell Expansion Medium. Activation of viable CD3+ T cells was assessed by CD25 expression using flow cytometry: the frequency of CD25 positive cells was 5.6 ± 2.4% (mean ± SD) on day 0, rising to 75.4 ± 13.8% (mean ± SD) after 3 days of culture.

For expansion studies, 1 x 10^6 EasySep™-isolated human T cells were stimulated with 25 μL of the activator in ImmunoCult™-XF T Cell Expansion Medium supplemented with 10 ng/mL Human Recombinant IL-2, with fresh medium and IL-2 added on days 3, 5, 7, and 10 over a 12-day culture period. No additional activator was added during this time, and results are reported as mean ± SD across 6 experiments with 3 donors.
