ImmunoCult-ACF Hu B Cell Expansion Supp

STEMCELL Technologies offers this serum-free and animal component-free culture supplement formulated to promote robust in vitro expansion of human peripheral blood B cells. The supplement enables B cell culture without serum, feeder cells, or specialized culture plates, and supports activation and expansion of human B cells along with their maturation to plasma cells.

The 50X concentrate is added to a base medium such as ImmunoCult™-XF B Cell Base Medium to prepare a complete serum-free medium for culturing and expanding human B cells, and is also available as part of the ImmunoCult™ Human B Cell Expansion Kit. It contains recombinant proteins, including CD40L and other cytokines, together with chemically defined components. The supplement is suited to applications in activation, cell culture, differentiation, expansion, and genome editing, with relevance to antibody development, immunology, and infectious disease research.

When used with the ImmunoCult™ Human B Cell Expansion Kit, B cells isolated from human PBMCs (leukopak) using the EasySep™ Human Pan-B Cell Enrichment Kit and seeded at 1 x 10^5 cells/well in 24-well tissue culture plates showed fold expansion ranging from 38- to 1190-fold at day 14 ± 1 day (n = 12 donors), with maturation to plasma cells/blasts indicated by CD138 and CD20 expression. Memory and naïve B cells isolated with the EasySep™ Human Memory B Cell Isolation Kit, seeded at 0.5 x 10^5 cells/well in 48-well plates and passaged every 2 - 4 days, were similarly supported.
