Description

From STEMCELL Technologies, this laboratory reagent is a ready-to-use, animal component-free culture medium intended to support the differentiation of human monocytes into dendritic cells (DCs) and their subsequent maturation. The serum-free, defined formulation is designed for use as part of an integrated workflow for generating mature DCs from isolated CD14+ monocytes, supporting downstream applications in cancer immunotherapy, vaccine, and infectious disease research.

According to the manufacturer's safety documentation, the product is a clear, red liquid that is not classified as a hazardous substance or mixture under the Globally Harmonized System (GHS) or the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200). It is stable under normal conditions and is not regulated for transport by DOT, TDG, MEX, ICAO, IATA, IMDG, RID, ADR, or ADN. Containers should be kept tightly closed in a dry, cool, and well-ventilated place. STEMCELL Technologies' quality management system is certified to ISO 13485, and the product is intended for research use only.

In the manufacturer's workflow, isolated monocytes are cultured in the medium with added differentiation supplement on day 0; the medium is refreshed with differentiation supplement on day 3, maturation supplement is added directly on day 5 without changing the medium, and mature DCs are harvested on day 7. When used with the matched ImmunoCult differentiation and maturation supplements, the average percentage of CD83-positive mature DCs at day 7 was 93 ± 5% with only 1 ± 1% of cells retaining the monocyte marker CD14 (mean ± SD, n=39). The yield of viable mature DCs corresponded to 45 ± 25% (mean ± SD, n=39). IL-12p70 concentrations in day-7 supernatant were 361 ± 81 pg/mL for mature DCs compared with 5 ± 2 pg/mL for immature DCs (mean ± SEM, n=27).
