Human T-Cell, Immune Regulator 1 ELISA Kit

This sandwich ELISA from Antibodies.com is intended for the quantitative determination of human TCIRG1 (T-Cell Immune Regulator 1) in serum and other biological fluids. The kit uses a 96-well microtiter plate pre-coated with an antibody specific for TCIRG1. Standards and test samples are added to the wells, and TCIRG1 present in each sample is captured by the immobilized antibody. After washing, a Biotinylated Anti-TCIRG1 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The absorbance is read at 450 nm in a colorimetric microplate reader and the TCIRG1 concentration is calculated by reference to the standard curve.

The assay has a reported sensitivity of 8.3 pg/ml and a range of 15.6–1000 pg/ml, with a total assay time of 4 hours 30 minutes. Sample recovery was characterised with n=5 for each matrix: serum showed a range of 80%–102% with an average of 91%, EDTA plasma showed 81%–101% with an average of 91%, and heparin plasma showed 80%–89% with an average of 84%. Dilution linearity was also reported across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma (n=5).

Kit components include a pre-coated microplate (12 x 8 well strips), lyophilized standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components are stored at +4°C or -20°C as instructed.
